A brain metastasis model for breast cancer using human embryonic stem cell-derived cerebral organoids

چکیده

Background: Breast cancer is a common cause of brain metastasis. Although breast has relatively high survival rates, its rate after metastasis to the lower. Conventional two-dimensional cell culture models and animal are widely used in metastatic research, these have tremendously contributed understanding this disease. However, some limitations, such as different physiological features genetic backgrounds. Methods: We established simple model using human pluripotent stem cell-derived cerebral organoids (COs)—in case, (BC-COs).Results: Using BC-CO model, we induced MDA-MB-231 cells into COs by co-culture with compared differences between adapted BC-COs non-adapted cells. Our results showed that proliferative capacity increased Additionally, expression levels endothelial-mesenchymal transition markers were significantly higher cells. Conclusion: conclude promotes also could be novel tool for research on cancer.

Generation of tissue-engineered small intestine using embryonic stem cell-derived human intestinal organoids

Short bowel syndrome (SBS) is characterized by poor nutrient absorption due to a deficit of healthy intestine. Current treatment practices rely on providing supportive medical therapy with parenteral nutrition; while life saving, such interventions are not curative and are still associated with significant co-morbidities. As approaches to lengthen remaining intestinal tissue have been met with ...

Cancer metastasis at a glance: A brief view of Breast Cancer Brain Metastasis

Cancer initiation and progression is a challenging scenario through developmental processes, which may be more complicated by occurrence of metastasis. Basically, diverse genetic alterations, including mutations, formation of truncated proteins and related pathways play key roles either in diagnosis or the most influential therapeutic purposes. ...

Modelling Zika Virus Infection of the Developing Human Brain In Vitro Using Stem Cell Derived Cerebral Organoids

The recent emergence of Zika virus (ZIKV) in susceptible populations has led to an abrupt increase in microcephaly and other neurodevelopmental conditions in newborn infants. While mosquitos are the main route of viral transmission, it has also been shown to spread via sexual contact and vertical mother-to-fetus transmission. In this latter case of transmission, due to the unique viral tropism ...

Drug Discovery via Human-Derived Stem Cell Organoids

Patient-derived cell lines and animal models have proven invaluable for the understanding of human intestinal diseases and for drug development although both inherently comprise disadvantages and caveats. Many genetically determined intestinal diseases occur in specific tissue microenvironments that are not adequately modeled by monolayer cell culture. Likewise, animal models incompletely recap...
